Study finds diabetes raises levels of proteins linked to Alzheimer's features

Growing evidence suggests that there may be a link between diabetes and Alzheimer's disease, but the physiological mechanisms by which diabetes impacts brain function and cognition are not fully understood. In a new study published in Aging Cell, researchers at the Salk Institute for Biological Studies show, for the first time, that diabetes enhances the development of aging features that may underlie early pathological events in Alzheimer's.

Specifically, the Salk team found increases in two hallmarks of Alzheimer's-accumulations of amyloid beta (Abeta) and tau protein-in the brains of diabetic mice, especially in cells surrounding blood vessels. Abeta, the misfolded peptide that is thought in part to cause Alzheimer's disease, aggregated inside astrocytes, star-shaped brain cells that, upon interaction with Abeta, release inflammatory molecules that can destroy neurons. Previously, this had not been shown in mouse models of type 1 diabetes (T1D).

"Our study supports and extends the links between diabetes, aging and Alzheimer's," says senior author Pamela Maher, a senior staff scientist in Salk's Laboratory of Cellular Neurobiology. "We show that type 1 diabetes increases vascular-associated amyloid beta buildup in the brain and causes accelerated brain aging."

The findings suggest that the neurovascular system may be a good candidate for new therapeutic targets to treat Alzheimer's in the early stages of the disease.

Alzheimer's and diabetes are two diseases that are increasing at an alarming rate within the U.S. population. Alzheimer's affects one in 10 Americans over 65 years of age and nearly 50 percent of those over 85. Similarly, more than 8 percent of Americans (approximately 26 million people) have diabetes, with the vast majority of those individuals being over 60.

Maher says her team is uncertain of the precise mechanism behind the increase in Abeta and tau in the mouse brain, but their data suggest that changes in astrocytes, as well as other pro-inflammatory processes and the bonding of proteins with sugar molecules (called non-enzymatic glycation), may contribute.

"Astrocytes play a key role in maintaining nerve cells in the brain," says lead study author Antonio Currais, a postdoctoral researcher at Salk. "Both chronic peripheral inflammation and increased non-enzymatic glycation are associated with diabetes, and these changes may act on the brain to alter astrocyte function, which eventually leads to Alzheimer's-like changes."

All nerve cells are closely connected to blood vessels, as they need nutrients---- especially glucose (sugar) and oxygen---- provided by the blood in order to function. Astrocytes facilitate the transfer of nutrients between blood vessels and cells. The buildup of Abeta at sites where astrocytes interact with blood vessels suggest that this could impair the transfer of nutrients. The type of Abeta localization seen in Maher's mouse models is also found in human Alzheimer's patients

To examine the contributions of diabetes to Alzheimer's-related pathology in the aged brain, the Salk researchers induced T1D in two sets of mouse models. One set, known as SAMP8 mice, undergo accelerated aging and develop early deterioration in learning and memory, as well as a number of brain alterations similar to those found in Alzheimer's. The other set, SAMR1 mice, which in this study came from the same gene pool as the SAMP8 mice, age normally.

Using these mice, Maher and her colleagues addressed how T1D interacts with age to contribute to Alzheimer's-related pathology. They showed that T1D elicits a wide range of pathological changes in the brains of both strains of mice, which are exacerbated by premature aging.

The Salk study is the first to show that these modifications are similar to those seen in old nondiabetic SAMP8 mice and to identify unique pathological changes, such as increases in markers for inflammation, in aged, T1D SAMP8 mice. Unlike most mouse studies of Alzheimer's, Maher's mice were not engineered to produce high levels of human Abeta or tau,so all of their observations came from naturally occurringAbeta and tau.

Once you're sitting on the freeway, a modern diesel--from Volkswagen or BMW, for example--will happily chug away at low engine speeds, sipping gas but offering enough torque in reserve should you need an extra burst of speed. Not only that, but low revs also means low noise--you're much more likely to hear wind and tire roar than you are any sounds from the engine. It's not a bad way to travel, and you'll be using very little fuel too.

WARSAW, Poland (AP) ? A tank used by Poland in its 1920 war against the Red Army was returned from Afghanistan where it was serving as a decoration at the defense ministry.

Defense Minister Tomasz Siemoniak said Friday the French-made FT-17 tank has "historic and sentimental value" for Poland.

According to historians it probably was captured by the Bolsheviks during the war, and later sent to Kabul as a gift.

It was spotted at the Defense Ministry in Kabul by an Afghan employee of Poland's embassy, according to Piotr Lukasiewicz, the Polish ambassador to Kabul.

After maintenance it will be displayed at the Polish Armed Forces Museum, Siemoniak said.

Europe has only three such tanks, the museum says. It is the first tank on tracks with front cabin and rear engine.

To demonstrate what the prototype can do, a camera observed a pair of workers helping each other drill into a loose metal plate. The camera measured the distances between the tools and work surface, while feedback from sensors on the workers' bodies tracked their movements. This taught the arms where to grab and how much force to apply, so it could then assist a lone worker to both hold the drill and secure the plate.

If you think the idea of free-roaming robotic arms holding power tools sounds alarming, you aren't alone. "If a robotic arm can do useful work, it can also hurt you badly," says Barrett. "Traditionally, people are kept far away from robot arms because the arms are dangerous. The concept of strapping robotic arms onto a person is terrifying," he says.

Parietti and Asada have tried to address some of those safety fears by building the arms from softer material. Flexible components in the robotic arm, called series elastic actuators - invented in the 1990s by Gill Pratt and Matt Williamson at MIT - mean that less damage will be done if the arms do lose control.

Dennis Hong at Virginia Tech in Blacksburg says that roboticists have spent the last 30 years attempting to make robots more springy and compliant, so they can work safely alongside humans. He says he has never come across robotic arms designed to be worn on the body.

The limbs were described at the Dynamic Systems and Control Conference in Florida last week. Funded by Boeing, their first use could be to help workers build aircraft. The broader goal, say the researchers, is for the limbs and their users to work seamlessly so that "humans may perceive them as part of their own bodies".

Why Are Our Brains So Ridiculously Big?

?What would you do with a brain if you had one?? Dorothy?s question to the Scarecrow in The Wizard of Oz elicited one of the movie?s most delightful songs, in which her straw-filled friend assured her that, among other things, he could ?think of things I?d never thunk before.? But the Scarecrow seemed to do quite well without one, thus avoiding the high energy costs of fueling and cooling a human brain?which, with an average volume of about 1,400 cubic centimeters, is humongous relative to our body size.

How did our brains get so big? Researchers have put forward a number of possible explanations over the years, but the one with the most staying power is an idea known as the social brain hypothesis. Its chief proponent, psychologist Robin Dunbar of Oxford University, has argued for the past two decades that the evolution of the human brain was driven by our increasingly complex social relationships. We required greater neural processing power so that we could keep track of who was doing what to whom.

Our expanded brains could have been practical for other things, of course, such as innovations in tool use and food gathering. Most researchers, including Dunbar, agree that these hypotheses are not mutually exclusive. Whatever the reasons for the very large human noggin, there is a lot of explaining to do, because big brains have a lot going against them.

The oversized Homo sapiens brain let us take over the planet, build cities, send space probes to Mars, and do all the other marvelous things that we humans are so proud of. But none of these things makes us much better at reproducing, and in terms of evolution, that?s really all that matters. It?s not so obvious why Darwinian natural selection should have favored the brain?s dramatic expansion given the huge costs. Although the human brain is only about 2 percent of total body weight, it siphons off about 20 percent of our total calorie intake; this overall percentage varies little whether we are engaged in hard mental tasks or just zoning out.

If having a large brain were all that advantageous, it seems that every animal would have one. And yet most species have been content, evolutionarily speaking, with relatively small ones. The brain of our closest living cousin, the chimpanzee, is less than a third the volume of ours, even though chimps weigh almost as much as humans.

Our primate lineage had a head start in evolving large brains, however, because most primates have brains that are larger than expected for their body size. The Encephalization Quotient is a measure of brain size relative to body size. The cat has an EQ of about 1, which is what is expected for its body size, while chimps have an EQ of 2.5 and humans nearly 7.5. Dolphins, no slouches when it comes to cognitive powers and complex social groups, have an EQ of more than 5, but rats and rabbits are way down on the scale at below 0.4.

Natural selection already had something to work with, if Dunbar?s hypothesis is correct, because many primates have complex social relationships. The best proxy for this social complexity, Dunbar argues, is the size of an animal?s social group. The social brain hypothesis really got cooking back in 1993, when Dunbar, along with anthropologist Leslie Aiello, crunched a bunch of numbers from the hominid fossil record and observations of living apes. The larger a species? group size, they found, the larger its brain?particularly the neocortex, the outer layers where most of the serious thinking goes on. They concluded that the correlation among group size, brain size, and neocortex size held pretty tightly throughout our lineage, from the australopithecines more than 3 million years ago to modern-day humans.

The average group size for today?s Homo sapiens, the pair concluded in a widely cited paper in Current Anthropology, is about 150 people?a figure often called Dunbar?s number, and which refers to the number of people with whom the average person can maintain close personal relationships. (The Australopithecus afarensis Lucy had a group size of 60 to 70, as do most living apes such as chimps, but Neanderthals were a close social-group match with modern humans.)

(AP) ? With the launch of Windows 8, buyers are about to discover a computing experience unlike anything they've seen before. Here's a guide to getting past some of the hurdles.

The main thing to know is that Windows 8 is designed especially for touch-screen computers, to make desktops and laptops work more like tablets. It is Microsoft's way of addressing the popularity of tablets, namely the iPad. But Windows 8 will work with mouse and keyboard shortcuts, too. It'll take some getting used to, though.

There are two versions of Windows 8, or more precisely, there's Windows 8 and there's Windows RT. They look the same, but they run on different processing chips. Windows 8 runs on standard chips from Intel and AMD and is the version you'd get if you're upgrading your home desktop or notebook PC. Windows RT is the version for light, small tablets and laptop-tablet hybrids.

Windows 8 will run programs written for older versions of Windows. Windows RT won't. It's limited to applications specifically written for it and available through Microsoft's store. (As a consolation, a version of Microsoft Office is included free on Windows RT devices).

Here are some tips on how to navigate the new Windows:

? When you start a Windows 8 machine, you're greeted with a screen that shows the time and a pretty picture. To get past it with a touch-screen device, swipe upwards with your finger from the bottom edge of the screen. If you have a keyboard, hit any key.

? Next, you'll see a mosaic of Live Tiles, each representing an application. Programs specifically written for Windows 8 will run in this new environment, which is unofficially nicknamed Metro. Each application fills the screen when you run it. Applications written for older Windows versions will open up in something that looks very much like the old Windows Desktop environment. You can switch back and forth between Metro and the new Desktop, though Microsoft wants people to eventually use only Metro.

? The Desktop screen lacks a Start button, so it's hard to start programs from there. Microsoft's idea is that users should learn to go to the Metro tiles to start programs or access settings, even if many programs, including some Windows utilities, will open up in Desktop. To get back to the tiled Start screen with a mouse or touchpad, move the mouse cursor to the top right corner of the screen, then swipe it down to the "Start" icon that appears. If you have a touch screen, reveal the Start icon by swiping in from the right edge of the screen.

? In the Desktop environment, you can glance at the Taskbar to see which Desktop programs are running. If you're a mouse or touchpad user in Metro and want to see what's running, you have to know this trick: Move the cursor into the top left corner of the screen, then drag it down along the left edge of the screen. If you have a touch screen, swipe in from the left edge, then quickly swipe back in.

? Neither environment will show you programs that are running in the other environment, but if you have a touch screen, swiping in from the left side of the screen lets you jump between open applications. The "Alt-Tab" combination does the same thing with a keyboard, in case you aren't using a touch screen.

? There are two versions of Internet Explorer, one for each environment. A Web page you open in one doesn't appear in the other, so if you're trying to find your way back to a page, it helps to remember which browser you were using.

? When using Metro on a touch screen, you close a program by first swiping your finger down from the top edge of the screen. That shrinks the window. Then you swipe your finger down to the bottom edge of the screen. Don't stray to the right or left edges of the screen, or the app will end up "docked" in a column along that edge. You can perform the same action with a mouse cursor by clicking and dragging from the top edge of the screen, but using the old "Alt-F4" command is easier.

? In the Desktop version of Internet Explorer, you can see at a glance which pages you have open in "tabs." In Metro, each Web page fills the screen, leaving no room for tabs.

To see which other pages are open on a touch-screen computer, you swipe your finger down from the top of the screen to reveal thumbnails of the other windows. Don't sweep too far, or you'll shrink the window instead.

If you're using a mouse in Metro, you right-click anywhere on the screen to reveal the tabs. Of course, this means right-clicking no longer does any of things it can be used for in previous versions of Windows, such as letting you open a link in a new tab.

? When Microsoft introduced Windows 95, some people thought it was amusing and counterintuitive that the procedure for shutting down the computer began with the "Start" button. In Windows 8, that incongruity is gone along with the Start button, but shutting down with a mouse or touchpad isn't obvious either. Move the cursor into the top right corner of the screen. A menu will pop out. Sweep down to the "Settings" button that appears, and click it. Then click "Power," then "Shut down." If you're on a touch screen, start by swiping in from the right edge of the screen, then tap "Settings."

In Portable Gaming, Nintendo 3DS XL Proves Bigger Is ... - AllThingsD

Bad news: My productivity went down ? way down ? this week. Good news: I?m getting pretty far in Super Mario Bros. 2.

There?s a reason I don?t allow myself to keep a gaming console and videogames in my house, and I was reminded of that as I logged some serious hours with the Nintendo 3DS XL.

This $200 portable gaming device offers a 3-D gaming experience without the need for special glasses.?It improves on the company?s previous model, the 3DS, with larger screens that make it easier to view and interact with the content, but it still has limitations, including low-resolution screens and limited 3-D viewing angles. Plus, the 3DS XL?s extra features, including the 3-D camera and Internet browser, aren?t that great.

Nintendo?s handheld also faces competition from Sony?s PlayStation Vita. That portable gaming system lacks 3-D, and costs $50 to $100 more than the 3DS XL, depending on if you get the Wi-Fi or cellular version, but it includes a higher-resolution screen and more processing power, which might appeal to the more serious gamer.

But for people like me who simply want a more robust gaming experience than what a smartphone offers, the 3DS XL is incredibly fun and addicting.

The 3DS XL is about the size of a small paperback book and weighs just under a pound. It features rounded edges, so it easily slips into a bag or jacket pocket, and feels comfortable to hold in both hands.

The clamshell device features dual displays, which Nintendo touts as being 90 percent larger than the previous Nintendo 3DS. The top non-touch display measures 4.88 inches diagonally, and the bottom touchscreen measures 4.18 inches, while the 3DS?s screens measure 3.53 inches and 3.02 inches, respectively. The PS Vita has a single five-inch touchscreen with a 960 x 544 pixel resolution.

When I borrowed a friend?s 3DS, my eyes strained a bit to take in all the details of a game. For example, I was playing Professor Layton and the Miracle Mask, a charming puzzle game that?s filled with great visuals as you travel through a fictional city called Monte d?Or. But it felt very cramped on the 3DS display, and I really had to focus to see some of the details of the background buildings, whereas the viewing experience on the 3DS XL was easier on the eyes.

That said, the larger bottom screen felt like the bigger luxury. The top display is used primarily to show you the game play, whereas the bottom screen allows you to control and select certain functions using touch. But that wasn?t always easy with the 3DS?s smaller screen, and I often had to use the included stylus. With the 3DS XL, I could just use my fingertip, which was more convenient.

Unfortunately, Nintendo didn?t increase the resolution of the screens along with the size. The top display has a resolution of 800 x 240 pixels, and the bottom has a resolution of 320 x 240 pixels. When blown up on the larger screens, the pixels are more noticeable, so images and text don?t look particularly sharp or smooth. The 3DS XL?s displays were still clear and bright enough that I could see them, but it?s an adjustment, coming from the higher-resolution screens found on today?s smartphones.

The 3-D effect is not the same 3-D you?d experience at the movies. Instead of objects flying off the screen, the 3DS XL creates an effect where parts of the image go deeper into the background while some items remain in the foreground to create the illusion of 3-D. It?s a pretty neat trick, though it works better on some games than others.?

For example, I thought it was better integrated on Super Mario Bros. 2 than on Professor Layton and the Miracle Mask. With the latter, there were certain scenes where the 3-D looked like a muddled mess. In those cases, there is a slider switch to the right of the top display that allows you to adjust the depth of the 3-D, or completely turn it off.

I was worried about getting a headache or feeling dizzy from the 3-D, but, surprisingly, I never felt sick the entire time I used the 3DS XL. It did take a few minutes for my eyes to adjust to the 3-D, and you do have to look at the screen straight on to see the effect. If you move your head or the handheld, you lose the 3-D image.?

Game play was a lot of fun. Some might wonder why not just play games on your smartphone, but having dedicated control buttons really makes a difference.

In the past, I downloaded several games to my iPhone, such as Mega Man II, but abandoned them after a few tries because trying to maneuver characters and complete actions using the onscreen touch controls was too hard. But the 3DS XL has a circle pad, a directional pad and four control buttons, and they all make game play so much easier, whether I was trying to run, jump or shoot.

This is part of the reason why I could play Super Mario Bros. 2 for so long. After about five hours of play (not continuous), the battery called ?game over? and needed a recharge.

Games are available as traditional cartridges, and range in price between $10 and $40, or you can purchase and download them over Wi-Fi right on the device, through the Nintendo eShop. I just about passed out from excitement when I saw The Legend of Zelda in the catalog, and readily paid the $5 to relive one of my favorite childhood videogames.

In addition to games, the eShop offers apps and 3-D videos, and downloaded content is saved on the preloaded four gigabyte SD card. Also, if you have one of Nintendo?s older DS, DSi or 3DS models, most games are compatible with the 3DS XL, but DS and DSi games will be displayed in 2-D.

The 3DS XL is more than just a gaming handheld. It?s equipped with three cameras ? the two on the front cover can capture 3-D images, and there?s one above the top display. But with a resolution of just 0.3 megapixels each, the picture quality is pretty poor.

The device also offers a basic Internet browser, but I would only use it out of desperation. Web sites, including Web-based email, don?t scale very well on the screens, and it requires a lot of zooming to be able to read text. There?s no Facebook or Twitter integration, but Nintendo includes its own social feature that lets you connect with other 3DS players.

If you?re looking to level up your game play beyond what?s offered by your smartphone, the Nintendo 3DS XL makes a fun companion.

NYPD busts potential cannibal cop

NEW YORK (AP) ? A city police officer was charged Thursday in a ghoulish plot to kidnap and torture women and then cook and eat their body parts.

Gilberto Valle sent numerous emails and other Internet communications about the torture and cannibalism scheme, according to a criminal complaint. He identified and catalogued at least 100 women on his computer, investigators said, but there was no information that anyone was harmed.

"I was thinking of tying her body onto some kind of apparatus ... cook her over low heat, keep her alive as long as possible," Valle allegedly wrote in one exchange in July, the complaint says.

In other online conversations, investigators said, Valle talked about the mechanics of fitting a woman's body into an oven (her legs would have to be bent), said he could make chloroform at home to knock a woman out and discussed how "tasty" one woman looked.

"Her days are numbered," he wrote, according to the complaint.

Valle was to appear in federal court in Manhattan on Thursday afternoon to face charges of kidnapping conspiracy and unauthorized use of law enforcement records. The name of his attorney was not immediately available, and no one answered the door to his home in a quiet, middle-class Queens neighborhood.

A search of Valle's computer found he created records of at least 100 women with their names, addresses and photos, the complaint says. Some of the information came from his unauthorized use of a law enforcement database, authorities said. He claimed, according to the complaint, that he knew many of them.

"The allegations in the complaint really need no description from us," said Mary E. Galligan, acting head of the FBI's New York office. "They speak for themselves. It would be an understatement merely to say Valle's own words and actions were shocking."

There was no immediate response to a message left with the NYPD on Thursday.

Valle met one potential victim over lunch, authorities said.

The complaint alleges that in February, Valle negotiated to kidnap another woman for someone else, writing, "$5,000 and she's all yours."

He told the buyer he was aspiring to be a professional kidnapper, authorities said.

"I think I would rather not get involved in the rape," according to the complaint. "You paid for her. She is all yours, and I don't want to be tempted the next time I abduct a girl."

It says he added: "I will really get off on knocking her out, tying up her hands and bare feet and gagging her. Then she will be stuffed into a large piece of luggage and wheeled out to my van."

Cellphone data revealed that Valle made calls on the block where the woman lives in March, the complaint says. An FBI agent interviewed the woman, who told them that she didn't know him well and was never in her home.

Valle, 28, lives in Queens. He had been assigned to a Manhattan precinct before his suspension on Wednesday.

His Facebook page cultivated the image of a very different man. Postings were filled with photos of a smiling wife, a baby girl and an English bulldog puppy named Dudley. A Maryland football and Yankees fan, Valle had more than 600 Facebook friends, including dozens of young women.

Valle respected his colleagues on the force, took the sergeant's exam and spoke out against Occupy Wall Street, cop killers and others who broke the law, according to the page. His current photo was a blue line, a sign of mourning for when an officer is killed, and expressed condolences for the family of a Nassau County officer who was shot to death this week.

"Keep Nassau County police in your prayers what a brutal week," he wrote earlier this week.

The page was taken down Thursday afternoon.

A man who identified himself as Valle's younger brother but did not give his name told reporters outside the officer's home that he was surprised by the arrest.

"You guys know more than I do," he said.
